On April 2, 2025, First Berlin Equity Research analysts Alexander Rihane and Christian Orquera initiated coverage of Rakovina Therapeutics Inc. (RKV:TSX.V) with a Buy rating and a price target of CA$0.40, highlighting the company's promising AI-accelerated pipeline of DNA Damage Response (DDR) compounds that position it for potential pharmaceutical partnership deals in the near term.
Rakovina is a pre-clinical stage biopharmaceutical company focused exclusively on developing next-generation anti-cancer compounds discovered through artificial intelligence (AI). The company's Deep Docking AI platform has been validated through real-world applications, including during the COVID-19 pandemic when it generated 1,000 potential drug candidates against SARS-CoV-2 in less than four weeks. Notably, a derivative of one of these compounds became the active ingredient in Pfizer's COVID drug Paxlovid. The platform's effectiveness has been further demonstrated by winning the international CACHE drug discovery competition twice in the last three years.
The company's lead program targets Poly ADP-Ribose Polymerase (PARP), a well-validated mechanism in oncology with first-generation PARP inhibitors marketed since 2014. Rakovina's kt-2000 next-generation PARP inhibitors are designed to address key limitations of current drugs, particularly poor brain penetration and high toxicity. The company aims to secure a licensing deal for this program in the second half of 2025 (First Berlin estimates: first half of 2026). Rakovina is also advancing an ATR inhibitor program (kt-5000), which is being prepared for out-licensing afterward (First Berlin estimates: second half of 2026).
The global PARP inhibitor market is projected to grow from US$4 billion in 2024 to US$16 billion by 2034, representing a compound annual growth rate of approximately 15%. The DDR space has seen strong deal activity, with around US$4.4 billion in total potential value across four major partnerships focused on ATR and PARP-1 inhibitors, demonstrating significant pharmaceutical industry interest in next-generation DDR therapeutics.
Upcoming catalysts include the presentation of two abstracts at the 2025 American Association for Cancer Research (AACR) Annual Meeting on April 25-30. These abstracts will present findings from in silico screening followed by in vitro and in vivo validation of the compounds' efficacy, selectivity, and pharmacokinetic properties — marking the first time Rakovina will publish real-world confirmation of their AI-generated compound data.
Key risks identified in the report include the company's small size and early-stage pipeline compared to peer DDR players, dependence on key staff, and the likelihood that Rakovina's compounds will not be first to market as competitor compounds are already in clinical testing. Financial risks are also significant, as failure to out-license the kt-2000 PARP inhibitor program would impact the company's ability to finance further development without substantial dilution.
The analysts value Rakovina at US$59.1 million based on a sum-of-the-parts methodology that applies a risk-adjusted net present value (rNPV) model for the lead drug candidates. With the share price at the time of the report at CA$0.10, the CA$0.40 target price represents a potential return of 300%.
